The incident happened a short time ago on the A628 Barnsley Road in Hoylandswaine.
Police say the road is completely closed and have asked motorists to avoid the area while the emergency services carry out their work.
No information has yet been released about the number of casualties or the severity of their injuries.
South Yorkshire Police have been asked to prove a statement on the incident.
