The Blades remain in the bottom three but the Yorkshire derby win over promotion chasing Leeds puts the pressure back on the teams above them.
It also dented Leeds’ hopes of an automatic promotion spot.
Sheffield United took the lead after the break when Leeds man Eric Lichaj scored an own goal and turned in Nick Montgomery’s effort.
Bjorn Helge Riise scored the second before Leeds sub Billy Paynter got a red card for violent conduct.
The Blades have scored fewer Championship goals this season than any other team and Leeds, who have scored more than any other team, came to the city as the in-form team.
But the Blades denied Leeds their first win at Bramall Lane since Brian Gayle’s famous own goal clinched the title for them in 1992.
