Yeovil v Barnsley pictures
Manager Simon Davey named all four of his summer signings in the starting 11 but at the break Darren Moore and Iain Hume came off.
Barnsley also gave a second-half trial to an unnamed trialist believed to be from the Czech Republic. He took the place of Diego Leon with the score at 1-0.
Yeovil's lively Danny Schofield created the first notable chance of the game squaring for Gavin Tomlin, but Barnsley's Mounir El Ha Haimour and Denis Souza managed to scramble the ball clear. A few minutes later a poor header let Schofield in, but his shot went wide.
On 40 minutes Paul Warne and Aidan Downes exchanged passes well down the left-hand side, before Warne delivered a cross met by Schofield, his shot was parried but only as far as Gavin Tomlin who slotted home.
Barnsley were struggling to create openings but a chance arrived when Jamal Campbell-Ryce's cross reached Stephen Foster's head. Keeper Corin Brown rescued the home side.
Tomlin then shot wide when it looked easier to score after some good play from Schofield and Warne.
A series of substitutions followed, including the introduction of Yeovil front pair Lloyd Owusu and Andy Kirk.
A superb free kick from Barnsley sub Brian Howard with 15 minutes to go was the closest the visitors came, the effort rocking the bar.
Yeovil made sure of the victory inside the last 10 minutes with a well taken goal from Owusu, slotting home with the aid of the upright.
Owusu and Kirk both had other opportunities but Barnsley stood firm.
